I'm pretty sure you have to unlock the big secrets in Konquest, cause you would have to earn them. And there are about 200 less koffins in the new Krypt.

Its from AT to TT now. Some of them you can only open with keys which you can find in the konquest mode.

There are 400 Koffins, as I recall. I believe there was 676 last time, so there is only 76 less than before, so thats not not to bad. Also it was said there will be more stuff you can use in these Koffins. Yes, they will only be unlocked though Konquest long after you completed the game. Hope that answers your questions.
