There's been several threads about this for various games, many people will tell you who they like in the games or who seems the neatest, but they don't understand that doesn't mean the best. What you are asking, is about cheapness/effectiveness, so friends, this thread is not about "Scorpion is the best in UMK3 because I say he is." or "Reptile is the best in MK2 because that was the game where he looked the best."
All characters have strengths and weaknesses, some more, some less. On a pound for pound basis however, these are the most official/accurate top tiers for Mortal Kombat 2D games:
MK1
Johnny Cage and Sonya
MK2
Mileena, Jax, Liu Kang, Kitana
MK3
Sub-zero, Kabal, Nightwolf, Jax, Kano, Robot Smoke
UMK3
Kabal, Human Smoke, Nightwolf, Kung Lao, Ermac, Kano, Robot Smoke, Jax
MKT
Noob Saibot, Kabal, Human Smoke, Baraka, Nightwolf, MK2 Kung Lao, Ermac, Rain, Kano, Robot Smoke, Jax

Somehow I still managed to misinterperate your post.
Playing against the computer is totally different than a person, my advice, find people to play against cause fighting the CPU is pointless.
Kitana is probably the best character to use against Kintaro and Shao Kahn.

It depends on which version of Kahn you're talking about. Even then forming tiers for fighting the CPU is tricky because everyone has their own little tricks and tactics and there's no real way to gauge which work best.
If you're talking MK II then I'd say Jax is the best against Kahn. You can just hit standing LK all day to beat him. You can do that with others, but his long reach for LK makes him the best at it. With MK3 I'd say eh, I dunno, Sheeva. It's really easy to just do the stomp over and over.

MKII Shao Khan has an easy pattern:
At the start of the round, keep jumping back until you get to the corner. Then just crouch and hold block. Wait for him to do his charging shoulder move, block it, and uppercut. Repeat until he dies.
Oh and every now and then he'll throw his green arrow projectile; just release block when he shoots it, so you don't take the chip damage. Then resume blocking and continue the pattern.
In MKII (at least the arcade version) this should work with any character.
